An EU summit tasked with solving the debt crisis and saving the euro has started in Brussels, with Germany and France already trying to smooth over their differences.

TONY JONES, PRESENTER: An EU summit tasked with solving the debt crisis and saving the euro has started in Brussels.

The German and French leaders have already held pre-summit talks aimed at smoothing over their differences.

Germany says the best way to strengthen the economy is by controlling national budgets, but France insists European governments must share debt obligations.

Italy is warning of a disaster if leaders at the summit don't find a way to co-operate.

MARIO MONTI, ITALIAN PRIME MINISTER: If the public opinion sees that bad signals, not good signals are coming back from the markets, well then people may be discouraged and political forces which say, "Let European integration, let the euro, let this or that large country go to hell," might prevail, which would be a disaster for the countries concerned and for the whole of the European Union.

TONY JONES: Left-wing protest groups are urging the leaders not to forget the poor in their deliberations. Plans on the take include a $160 billion stimulus package and the creation of a European treasury with power over national budgets.

Obama prepping legion of lawyers for election

OLYMPIA, Wash. (AP) ? President Barack Obama's campaign has recruited a legion of lawyers to be on standby for this year's election as legal disputes surrounding the voting process escalate.

Thousands of attorneys and support staffers have agreed to aid in the effort, providing a mass of legal support that appears to be unrivaled by Republicans or precedent. Obama's campaign says it is particularly concerned about the implementation of new voter ID laws across the country, the possibility of anti-fraud activists challenging legitimate voters and the handling of voter registrations in the most competitive states.

Republicans are building their own legal teams for the election. They say they're focused on preventing fraud ? making sure people don't vote unless they're eligible ? rather than turning away qualified voters.

Since the disputed 2000 presidential election, both parties have increasingly concentrated on building legal teams ? including high-priced lawyers who are well-known in political circles ? for the Election Day run-up. The Bush-Gore election demonstrated to both sides the importance of every vote and the fact that the rules for voting and counting might actually determine the outcome. The Florida count in 2000 was decided by just 537 votes and ultimately landed in the Supreme Court.

This year in that state alone, Obama and his Democratic allies are poised to have thousands of lawyers ready for the election and hope to have more than the 5,800 attorneys available four years ago. That figure was nearly twice the 3,200 lawyers the Democrats had at their disposal in 2004.

Romney has been organizing his own legal help for the election. Campaign attorney Ben Ginsberg did not provide numbers but said the campaign has been gratified by the "overwhelming number of attorneys who have volunteered to assist."

"We will have enough lawyers to handle all situations that arise," he said.

The GOP doesn't necessarily need to have a numerical counterweight to Obama's attorneys; the 2000 election showed that experienced, connected lawyers on either side can be effective in court.

Former White House counsel Robert Bauer, who is organizing the Obama campaign's legal deployment, said there is great concern this year because he believes GOP leaders around the county have pursued new laws to impede the right to vote.

"The Republican Party and their allies have mapped out their vote suppression campaign as a response to our success in 2008 with grass-roots organization and successful turnout," Bauer said. "This is their response to defeat: changing the rules of participation so that fewer participate."

Several states with Republican leaders have recently pursued changes that could make voting more difficult, including key states such as Florida and Ohio, despite objections from voting rights groups that believe that the laws could suppress votes from low-income and minority blocs.

Republicans dispute that the laws are political, pointing to cases of election fraud and arguing that measures like those requiring voters to show identification are simply common sense. Pennsylvania's Republican House majority leader, Mike Turzai, however, told GOP supporters over the weekend that the state's new ID law "is going to allow Governor Romney to win the state of Pennsylvania."

Independent from the Romney team, a conservative group is prepping an Election Day team of its own to combat possible fraud.

Catherine Engelbrecht, president and founder of True the Vote, said the organization hopes to train and mobilize up to one million volunteers this year, many of them to serve poll watchers. One of the group's main initiatives is to "aggressively pursue fraud reports."

"Being a poll watcher is an age-old tradition and we're fortunate that so many volunteers are ready and willing to take a day off, learn what they need to know and help out at the polls," Engelbrecht said. True the Vote already has thousands signed up to help and had 500 trained election workers monitoring the Wisconsin recall vote earlier this month.

"They serve as volunteer guardians of the republic, to ensure that procedures at the polls are in keeping with state law," she said.

It's one of the efforts that have Obama's team fretting. The Democrats fear that anti-fraud activity could get out of hand, with vigilante poll watchers targeting and intimidating voters who may not know their rights.

"We will have the strategy and the resources to address the threat and protect the voter," Bauer said.

The Obama-aligned attorneys, most of whom are not election experts by trade, undergo training and have materials to show them how to help at the polls on Election Day.

Charles Lichtman, who is helping advise the effort in Florida this year after leading it in the last two cycles, first created the Florida Democratic Lawyers Council after the 2000 election, vowing that there would never be a repeat of that disputed vote. He contends Democrat Al Gore would have won the presidency over Republican George W. Bush if a similar legal infrastructure had been in place then.

Lichtman's efforts have since been replicated for other states. He said that is vital to provide voter protection.

"My experience has been that, in every election, the other side has taken drastic measures to try to suppress the vote," Lichtman said. The volunteer organization has not been involved in the 2012 legal disputes so far, though they are monitoring the developments.

Four years ago, the teams of lawyers organized by Obama and Republican candidate John McCain in 2008 went largely unused since the election wasn't very close.

But this year may be different given all the changes to voting laws ? and the closeness of the race in recent polling.

The states with the strictest ID laws require voters to show photo identification before casting ballots. If they don't have proper identification or fail to bring it, they can cast a provisional ballot but must later go to meet with state elections administrators to sort things out before the ballot is counted.

Voting groups see a variety of potential problems, such as how voters are informed of the rule changes, how poll workers handle voters who fail to bring IDs and whether voters are provided adequate notice of the steps they need to take after casting an absentee ballot.

About 30 states have some form of an ID law, with varying methods of implementation.

Legal challenges typically start coming in the weeks before the election, but "litigation has started coming sooner and more vociferously" this year, says Edward Foley, an elections law expert with Ohio State University. That includes lawsuits surrounding Florida's plan to purge ineligible voters from the rolls.

Foley said. "We're in an era of increased litigiousness over the voting process."

He said lawsuits after Election Day may occur only if votes in a battleground state are within the "margin of litigation." That would probably be a difference of just hundreds of votes, a result that would be rare.

ScienceDaily (June 27, 2012) ? A new study confirms that doing short journeys on foot rather than taking the car or motorbike would avoid the death of 108 men and 79 women a year in Catalonia alone. This would imply annual savings of more than 200 million euros.

Researchers from the World Health Organisation (WHO) and the Barcelona Public Health Agency (ASPB), headed by Catherine P?rez from the Healthcare Information Systems Service, have estimated the yearly economic benefits from a reduction in death rates by substituting at least one short vehicle journey for a walk.

The authors conducted a cross-sectional study based on Catalonia's Mobility Survey performed by the Generalitat and the Metropolitan Transport Agency in 2006 which documents the journeys of more than one hundred thousand people.

Then, 80,552 individuals over 17 years of age who did at least one journey were selected and researchers calculated the number of men and women who did not meet daily physical exercise recommendations but travelled by car or motorbike for journeys up to five minutes long.

In order to calculate the annual economic benefits, the 'Health Economic Evaluation Tool (HEAT)' was used. It was designed by the WHO and estimates the benefits of reduced mortality by increasing physical exercise.

For their economic estimations, as set out by HEAT, the researchers took in account certain parameters: 150 minutes walked each week, a relative risk of 0.78 for all deaths linked to walking half an hour a day, a mortality rate in Spain of 21,349 deaths per 100,000 people between the ages of 20 and 49 years and a statistical value of a person's life of ?1,300,000 according to the "willingness to pay" methodology.

This study has been published in the 'European Journal of Public Health' and its results reveal that 77.2% of men and 67.7% of women in Catalonia do not reach recommended daily amounts of physical exercise when walking.

"However, 15.6% and 13.9% respectively would reach recommendations if they were to substitute at least one short, 5 minute vehicle journey," as explained by Marta Olabarria, lead author of the study.

Through the Global Strategy on Diet Physical Activity and Health, the WHO states that it is necessary to do at least 150 minutes of moderate aerobic exercise per week. Walking to get around is considered a moderate physical activity and is perfect to meet exercise recommendations.

You can install a 2.5 inch SATA drive but you have to supply it yourself since the device ships diskless.? Internal maximum storage capacity is 1.5TB. You can attach USB external drives formatted with FAT32, NTFS, HFS, EXT2, EXT3, EXT4, or XFS. The device also supports USB printers.

The internal SATA disk and the external eSATA drive being attached can be configured with RAID 0 or RAID 1 to expand capacity.

Setup
The setup process leaves room for improvement. First, there isn't sufficient documentation about installing and configuring the SATA drive.? There's no setup software, so you have to rely on the hard copy Quick Install guide that the NAS ships with, which has somewhat confusing instructions.

After installing the HDD, the guide instructs connecting the NAS to your network. The next step is to mount a partition from a newly installed SATA or USB drive from the Disk Manager. Although the guide does not specify how to get to Disk Manager, I found Disk Manager located within the Web-based management software. You have to login to the Web portal first, so the instructions seem a bit backward.

The web-based management portal is available at www.myakitio.com. To access, you have to enter the MAC address of the device and a username and password; all of which are located on a sticker at the bottom of the NAS. You can register the MyCloud Mini and give it an actual hostname, rather than use the MAC address every time to login.
The Web interface, which is called iSharing, seems to have an issue handling a fresh install of a new drive. When I first accessed the management interface home page, I saw a spinning cursor. When the spinning stopped, I got the message, "Get data error."

When I clicked off the message, another window displayed that the system was running without a home partition and needed to be mounted. I clicked "Mount," but the system did not detect any drives. I decided to manually start up Disk Manager.

Disk Manager gives three options to configure disks: Auto, Advanced, and Expert. The latter two allow for manual configuration and with Auto, the system creates a suitable partition and formats the drives. Auto is the recommended option.? I selected Auto and again, got the circling cursor, then a message on-screen: "Completed."

There was no further information about viewing the drives, or what was actually done to them. I clicked Disk Manager again and once more, got the screen which lets you choose from Auto, Advanced, and Expert drive setup.

Reaching a point of exasperation, I selected "Expert" as a setup option, and could see my SATA drive was configured with a home partition formatted with XFS as well as a swap partition. The setup process is circuitous, unintuitive, and could flow better.

The first time Jolene Rudell fainted, she assumed that the stress of being in medical school had gotten to her. Then, two weeks later, she lost consciousness again.

Blood tests showed Ms. Rudell?s red blood cell count and iron level were dangerously low. But she is a hearty eater (and a carnivore), and her physician pointed to another possible culprit: a popular drug used by millions of Americans like Ms. Rudell to prevent gastroesophageal acid reflux, or severe heartburn.

Long-term use of the drugs, called proton pump inhibitors, or P.P.I.?s, can make it difficult to absorb some nutrients. Ms. Rudell, 33, has been taking these medications on and off for nearly a decade. Her doctor treated her anemia with high doses of iron, and recommended she try to manage without a P.P.I., but that?s been difficult, she said. ?I?m hoping I?ll get off the P.P.I. after I complete my residency training,? she said, ?but that?s still several years away.?

As many as four in 10 Americans have symptoms of gastroesophageal reflux disease, or GERD, and many depend on P.P.I.?s like Prilosec, Prevacid and Nexium to reduce stomach acid. These are the third highest-selling class of drugs in the United States, after antipsychotics and statins, with more than 100 million prescriptions and $13.9 billion in sales in 2010, in addition to over-the-counter sales.

But in recent years, the Food and Drug Administration has issued numerous warnings about P.P.I.?s, saying long-term use and high doses have been associated with an increased risk of bone fractures and infection with a bacterium called Clostridium difficile that can be especially dangerous to elderly patients. In a recent paper, experts recommended that older adults use the drugs only ?for the shortest duration possible.?

Studies have shown long-term P.P.I. use may reduce the absorption of important nutrients, vitamins and minerals, including magnesium, calcium and vitamin B12, and might reduce the effectiveness of other medications, with the F.D.A. warning that taking Prilosec together with the anticlotting agent clopidogrel (Plavix) can weaken the protective effect (of clopidogrel) for heart patients.

Other research has found that people taking P.P.I.?s are at increased risk of developing pneumonia; one study even linked use of the drug to weight gain.

Drug company officials dismiss such reports, saying that they do not prove the P.P.I.?s are the cause of the problems and that many P.P.I. users are older adults who are susceptible to infections and more likely to sustain fractures and have nutritional deficits.

But while using the drugs for short periods may not be problematic, they tend to breed dependency, experts say, leading patients to take them for far longer than the recommended 8 to 12 weeks; some stay on them for life. Many hospitals have been starting patients on P.P.I.?s as a matter of routine, to prevent stress ulcers, then discharging them with instructions to continue the medication at home. Dr. Charlie Baum, head of U.S. Medical Affairs for Takeda Pharmaceuticals North America Inc., said its P.P.I. Dexilant is safe when used according to the prescribed indication of up to six months for maintenance, though many physicians prescribe it for longer.

?Studies have shown that once you?re on them, it?s hard to stop taking them,? said Dr. Shoshana J. Herzig of Beth Israel Deaconess Medical Center in Boston. ?It?s almost like an addiction.?

P.P.I.?s work by blocking the production of acid in the stomach, but the body reacts by overcompensating and, she said, ?revving up production? of acid-making cells. ?You get excess growth of those cells in the stomach, so when you unblock production, you have more of the acid-making machinery,? she said.

Moreover, proton pump inhibitors have not been the wonder drugs that experts had hoped for. More widespread treatment of GERD has not reduced the incidence of esophageal cancers. Squamous cell carcinoma, which is associated with smoking, has declined, but esophageal adenocarcinomas, which are associated with GERD, have increased 350 percent since 1970.

?When people take P.P.I.?s, they haven?t cured the problem of reflux,? said Dr. Joseph Stubbs, an internist in Albany, Ga., and a former president of the American College of Physicians. ?They?ve just controlled the symptoms.?

And P.P.I.?s provide a way for people to avoid making difficult lifestyle changes, like losing weight or cutting out the foods that cause heartburn, he said. ?People have found, ?I can keep eating what I want to eat, and take this and I?m doing fine,??? he said. ?We?re starting to see that if you do that, you can run into some risky side effects.?

Many patients may be on the drugs for no good medical reason, at huge cost to the health care system, said Dr. Joel J. Heidelbaugh, a family medicine doctor in Ann Arbor, Mich. When he reviewed medical records of almost 1,000 patients on P.P.I.?s at an outpatient Veterans Affairs clinic in Ann Arbor, he found that only one-third had a diagnosis that justified the drugs. The others seemed to have been given the medications ?just in case.?

?We put people on P.P.I.?s, and we ignore the fact that we were designed to have acid in our stomach,? said Dr. Greg Plotnikoff, a physician who specializes in integrative therapy at the Penny George Institute for Health and Healing in Minneapolis.

Stomach acid is needed to break down food and absorb nutrients, he said, as well as for proper functioning of the gallbladder and pancreas. Long-term of use of P.P.I.?s may interfere with these processes, he noted. And suppression of stomach acid, which kills bacteria and other microbes, may make people more susceptible to infections, like C. difficile.

Taking P.P.I.?s, Dr. Plotnikoff said, ?changes the ecology of the gut and actually allows overgrowth of some things that normally would be kept under control.?

Stomach acid also stimulates coughing, which helps clear the lungs. Some experts think this is why some patients, especially those who are frail and elderly, face an increased risk of pneumonia if they take P.P.I.?s.

But many leading gastroenterologists are convinced that the benefits of the drugs outweigh their risks. They say the drugs prevent serious complications of GERD, like esophageal and stomach ulcers and peptic strictures, which occur when inflammations causes the lower end of the esophagus to narrow.

The studies that detected higher risks among patients on P.P.I.?s ?are statistical analyses of very large patient populations. But how does that relate to you, as one person taking the drug?? said Dr. Donald O. Castell, director of esophageal disorders at the Medical University of South Carolina and an author of the American College of Gastroenterology?s practice guidelines for GERD, who has financial relationships with drug companies that make P.P.I.?s. He added, ?You don?t want to throw the baby out with the bathwater.?

Most physicians think that GERD is a side effect of the obesity epidemic, and that lifestyle changes could ameliorate heartburn for many.

?If we took 100 people with reflux and got them to rigidly follow the lifestyle recommendations, 90 wouldn?t need any medication,? Dr. Castell said. ?But good luck getting them to do that.?

Parenting is not how i imagined it! - Family - Support Groups

Im wondering if anyone else had these huge ideals of what kind of parent they would be-and then never became that type of parent.

i always try to do as much as i can for my kids-worry about them-but i know i spend so much time worrying about the future-that i dont enjoy present. i also am not the cool, fun mom i wish to be. im not supermom.im am alot like my own mom lol which isnt a bad thing-but i never thought i'd be like her. I use to get so annoyed w. my mom when she worried or was nervous person--or when she was busy doing 10 things at once--and now i see myself doing the exact same thing. i always wanted to shield my children from pain, dysfunction etc--but this last yr w. issues w. their dad-being hurt/sick-they have seen me upset-and likely they were confused/upset themselves at the situation..

one thing i finally realized though that my kids are very resillent and egocentric. their under 6 so its normal for them to be self focused. They arent selfish-have empathy for others-but they view things only from their persepctive-and how it affects them. i felt so awful-that i was in hospital -then getting help=not w/them--thought they hate me or feel abandoned. But now that im home w. them- i realize my kids werent devestated. they were w. family-didnt really realize exactly what happened-and they were more worried about little things in their life-school-friends-their toys-my daughter was worried about not being able to watch dora in a new place lol. She didnt realize Dora could be viewed on any tv that got the channel. So i realize they will likely be ok-even if i cant provide them the perfect life, free of drama/sad things etc. I hope small stresses in life-will teach them that stuff happens but we can overcome it as a family.

i use to be a stay at home mother-but went back to school. Now im in school/working part time-and i was so worried that my kids would be sad w.me not home and-id be a bad mom. my biggest fear is that im a bad mom. but oddly somehow it all worked out. they like day camp-i still put them first-we have lots of time together -so it doesnt matter that im not home w. them. plus they are hitting the ages were they have friends-outside activities like classes-so they like socializing-and telling me about their day.

what i realize today is there is no one right way to parent-and most of us do our best and make it work. kids are resilient-and as long as they know we love them-and are securely attached to us--they will be fine.

im taking a lifespan developmental psych class over summer online--and few of my fellow students hotly debated whether parent shouldnt cuddle too much there kids-or if the new trendy attachment parenting is right way. We were suppose to be discussing attachment bw infant and caregiver==but some how it lead to parenting styles-and what is the right way.---i didnt mean it offensively to them--but who cares. its really a personal choice of a parent. I cuddled my kids, but eventually put them in a crib too--i was a teenager-so needed my moms/husband to help to learn how to be a mom-and was so overwhelmed that i did whatever would calm my son. my first was difficult-so he may have been cuddled alot-but he is secure, normal-not overly attached 6 yr old who is happy. First 5 mths of his life-he slept more w. me or his dad then in bassinet or crib-but he survived=learned to self soooth and is ok. Even w. breast feeding-health wise maybe little better-but doesnt matter-u will still have an ok happy kid-if u dont breast feed. I breast feed-but i would never judge a woman who chooses not too-or couldnt, what matters is parents caring-involved w. there kids. the kids who sadly are insecuely attached to parents or not all attached were neglected, abused, and some dont even have loved ones who care about them.

SO i am trying very hard not to over worry or get down on myself if i cant be the perfect parent-bc i realize there is no perfect parent.

When drama subsided, I found my sons old knight custome=she went as a "knight" w. her brother's light up sword too-and had a blast==and said she didnt want to be a princess-she wanted to be a knight-so i realize things just work out-we all have to be resilient in this world.

sorry for tangent. i can be so hard on myself-and worry im not a good parent. but it just hit me-that when we worry-love our kids-and try our hardest--even if we make mistakes--we are ok--even good parents--and odds are our children are ok and happy =)

On aside note-ideal things i always wanted to do w. my kids-is over summer do car trip-travel-learn about different states. so i will try to do that some day. Maybe even this summer it could be a mini car trip/vacation. Main thing is im going to learn to be ok doing what works for me and my family, and stop feeling bad that im not super mom---or do things the same way everyone else does.
I also worry my kids will just value material things-so i want to start getting them involved in helping others-maybe get them involved in giving a toy to the poor or something like that.

Tropical Storm Debby drenches Florida

TAMPA, Fla. (AP) ? Practically parked off Florida's Gulf Coast since the weekend, Tropical Storm Debby raked the Tampa Bay area with high wind and heavy rain Monday in a drenching that could top 2 feet over the next few days and trigger widespread flooding.

At least one person was killed Sunday by a tornado spun off by Debby in Florida, and Alabama authorities searched for a man who disappeared in the rough surf.

An estimated 35,000 homes and businesses lost electricity. But as of midafternoon, the slow-moving storm had caused only scattered damage, including flooding in some low-lying areas.

The bridge leading to St. George Island, a vacation spot along the Florida Panhandle, was closed to everyone except residents, renters and business owners to keep looters out. The island had no power, and palm trees had been blown down, but roads were passable.

"Most true islanders are hanging in there because they know that you may or may not be able to get back to your home when you need to," said David Walker, an island resident having a beer at Eddy Teach's bar. He said he had been through many storms on the island and Debby was on the weaker end of the scale.

Gov. Rick Scott declared a statewide emergency, allowing authorities to put laws against price-gouging into effect and override bureaucratic hurdles to deal with the storm.

By late afternoon, Debby was in the Gulf of Mexico, 30 miles southwest of Apalachicola, with sustained winds around 45 mph.

A tropical storm warning remained in effect for the coastal counties of west-central and southwest Florida, forecasters said Monday evening.

Forecasters said it would crawl to the northeast, come ashore along Florida's northwestern coast on Wednesday and track slowly across the state, exiting along the Atlantic Coast by Saturday morning and losing steam along the way.

Parts of northern Florida could get 10 to 15 inches of rain, and some spots as much as 25 inches, as the storm wrings itself out, forecasters said.

"The widespread flooding is the biggest concern," said Florida Emergency Operations Center spokeswoman Julie Roberts. "It's a concern that Debby is going to be around for the next couple of days, and while it sits there, it's going to continue to drop rain. The longer it sits, the more rain we get."

High winds and the threat of flooding forced the closing of an interstate highway bridge that spans Tampa Bay and links St. Petersburg with areas to the southwest.

Monday evening, the state announced the closing of the Howard Frankland bridge that connects Tampa, including the region's major airport, and St. Petersburg. The eight-lane bridge carries Interstate 275 over Tampa Bay. Traffic was being diverted to an alternative span.

People in several sparsely populated counties near the crook of Florida's elbow were urged to leave low-lying neighborhoods because of the danger of flooding. Shelters opened in some places.

On St. Pete Beach in the Tampa Bay area, surfers enjoyed the large waves in the Gulf, which is usually so calm the water looks like glass. Residents cleaned up debris in yards and streets from a possible tornado Sunday.

"The wind picked up so bad. It's very, very scary. I ran into the closet underneath the hallway stairs," said Ann Garrison, who has lived on the barrier island for 20 years but has never seen such strong winds. She said that when she came back out after just a few minutes, "the fence was gone, and it was in the middle of the yard."

Nearby, a likely tornado ripped the roof off a marina and an apartment complex and knocked down fences, trees and signs.

Kourosh Bakhtiarian's yard was flooded. He said people were driving around the neighborhood to gawk at the damage, and he complained that police hadn't closed off the streets.

"We have a lot of visitors from outside of this area. They just want to see exactly where the disaster is. I mean, this is not the happiest time," he said.

On St. George Island, many businesses were closed, but Eddy Teach's bar had a few customers and used a generator to keep beer and food cold.

"The tourists cleared out. It's not a good thing and hurts the economy during a week in peak season," said Patrick Sparks, a manager at the bar. He scoffed at the storm, which was well below the 74 mph threshold for a hurricane: "It's a little rash to send everyone home."

Facebook names 1st woman, Sandberg, to board

(AP) ? Facebook has named its No. 2 executive, Chief Operating Officer Sheryl Sandberg, to its board of directors.

Sandberg, who joined Facebook from Google in 2008, is the first woman on Facebook's board of directors. People had called for the company to add a woman to its board ahead of its initial public offering of stock in mid-May.

Sandberg, 42, joins CEO Mark Zuckerberg, Netflix chief Reed Hastings and entrepreneurs Marc Andreessen, James Breyer and Peter Thiel. Also on the board are Donald Graham, CEO of The Washington Post Co., and Erskine Bowles, president emeritus of the University of North Carolina system.

Sandberg's appointment is somewhat symbolic. Zuckerberg controls Facebook and its board through more than half of its voting stock.

RUB researcher and Japanese colleagues report in the Journal of Biological Chemistry

How single cell organisms like bacteria manage to react to their environment is not yet completely understood. Together with colleagues from Japan, Dr. Samir El-Mashtoly from the RUB Department of Biophysics, led by Prof. Dr. Klaus Gerwert, has gained new insights into the molecular interactions during aerotaxis of Bacillus subtilis, i.e., the dependence of the movement direction on the oxygen concentration in the environment. The research team investigated the conformational changes within the protein HemAT. Via a signal transduction chain, this protein sends a command to the flagellar motor which controls the movement direction. They report in the Journal of Biological Chemistry.

Signal transduction chain

The signal transduction chain starts with binding of oxygen to HemAT's heme domain, which is also known from haemoglobin in the red blood cells and is called the sensor domain of HemAT. Oxygen binding leads to a conformational change in the sensor domain. This in turn provokes several further conformational changes within HemAT that finally affect the signalling domain of the protein. The signalling domain then transmits the information about a rise in oxygen concentration to other proteins within the cell. These proteins forward the message to the motor of the flagellum. The research team investigated how the information travels from the sensor domain of HemAT to its signalling domain.

Protein helices forward the information

For that purpose, Dr. El-Mashtoly used the time-resolved ultraviolet resonance Raman spectroscopic facilities in the Picobiology Institute in Japan. This method provides, for instance, structural information about the conformation of the protein and hydrogen bonding interactions on a nanosecond to microsecond time scale. The results suggest that the conformational change in the sensor domain, i.e., the heme structure, induces the displacement of two protein helices within HemAT. This displacement affects another helix which is continuous with the structure of the signalling domain. Due to a series of conformational changes, the information about oxygen binding thus reaches the signalling domain of the protein.
